Structure-Function Differences Among Type I Restriction Endonuclease


The stoichiometry of EcoR124I allows HsdS to access the DNA substrate independent of the presence of SAM

The fact that EcoR124I has been shown to be capable of both DNA cleavage and ATPase activity independent of the presence of SAM suggests that the enzyme is functionally different from EcoKI. Initial determination of molecular weight and stoichiometry suggest that the enzyme has a stoichiometry of R1M2S1. Therefore, we propose that EcoR124I has a major structure-function difference from EcoKI. This difference lies in the fact that only one HsdR subunit is present in the complex, and this allows DNA binding to occur without a requirement for SAM as an allosteric effector.
