Translocation follows the helical thread of the DNA

During DNA translocation Type I R-M enzymes bind to the adjacent DNA in cis and then move this DNA through the enzyme complex following the helical thread of the DNA.  This results in positive and negative supercoiling, in front or behind the enzyme, on circular DNA (Figure 6a).  These twisted structures can be seen in electron micrographs of translocated circular DNA (Figure 6b).
